Mathematical Formula & Assumptions

Cells / mL = Average cells per square × Dilution factor × 10^4
Key Assumptions & Rules:
  • Chamber volume factor is default 10^4, assuming a standard depth of 0.1 mm and square grid dimensions of 1 mm x 1 mm (0.1 mm³ per square).
  • Viability is computed using trypan blue exclusions: viable cells divided by total counted cells.
  • Optimal loading limits per square: 20 to 100 cells. Counts outside this range trigger density advisory messages.
